چکیده

Recent studies have shown that cytokine/cytokine receptor systems affect leukemic cell biology and clinical behavior of leukemic patients. Among various cytokine receptors, only interleukin-2 receptor α-chain (IL-2Rα, CD25) expression predicts a poor prognosis of patients (≤ 60 years old) with acute myeloid leukemia (AML). Even in prognostic analyses including unfavorable surface markers and cytogenetics, IL-2Rα is recognized to be an independent adverse indicator in such patients. Assessment of the presence of IL-2Rα should be essential to make therapeutic risk classification of AML patients in a future. However, as the lack of IL-2 responsiveness is observed in AML cells, further investigations are required to clarify the mechanism why IL-2Rα(+) AML patients have a dismal clinical outcome.

Prognostic Relevance of Cytokine Receptor Expression in Acute Myeloid Leukemia: Interleukin-2 Receptor α-Chain (CD25) Expression Predicts a Poor Prognosis

A variety of cytokine/cytokine receptor systems affect the biological behavior of acute leukemia cells. However, little is known about the clinical relevance of cytokine receptor expression in acute myeloid leukemia (AML). We quantitatively examined the expression of interleukin-2 receptor α-chain (IL-2Rα, also known as CD25), IL-2Rβ, IL-3Rα, IL-4Rα, IL-5Rα, IL-6Rα, IL-7Rα, the common β-chain (...

Expression of CD25 antigen on CD34+ cells is an independent predictor of outcome in late-stage MDS patients treated with azacitidine

The expression of CD25 antigen, the a-chain of the interleukin-2 receptor (IL-2Ra), has been repeatedly shown to be associated with poor outcome in adults with either acute myelogenous (AML) or lymphoblastic leukemia. To explain this finding mechanistically, it has been hypothesized that CD25þ AML blasts are enriched in chemoresistant leukemia stem cells (LSCs), as they bear an LSC-like molecul...

Interleukin-23 Receptor Gene Variants in Acute Lymphoblastic Leukemia and Their Relation to Prognostic Factors

Background: Interleukin (IL)-23 has an important role in tumor immune regulation. Objective: To investigate the possible association of interleukin-23 receptor (IL23R) gene variants rs1884444, rs10889677 and rs11209026 with development of acute lymphoblastic leukemia (ALL). Methods: The IL23R variants were studied in 164 ALL patients and compared to 175 healthy controls by polymerase chain reac...

A Leukemia-Associated CD34/CD123/CD25/CD99+ Immunophenotype Identifies FLT3-Mutated Clones in Acute Myeloid Leukemia.

PURPOSE We evaluated leukemia-associated immunophenotypes (LAIP) and their correlation with fms-like tyrosine kinase 3 (FLT3) and nucleophosmin (NPM1) gene mutational status in order to contribute a better identification of patients at highest risk of relapse in acute myeloid leukemia (AML). EXPERIMENTAL DESIGN Bone marrow samples from 132 patients with AML were analyzed by nine-color multipa...

A CD25− Positive Population of Activated B1 Cells Expresses LIFR and Responds to LIF

B1 B cells defend against infectious microorganisms by spontaneous secretion of broadly reactive "natural" immunoglobulin that appears in the absence of immunization. Among many distinguishing characteristics, B1 B cells display evidence of activation that includes phosphorylated STAT3. In order to identify the origin of pSTAT3 we examined interleukin-2 receptor (IL-2R) expression on B1 cells. ...
